چکیده

Abstract Background Colorectal cancer (CRC) is related to gut microbiota dysbiosis, especially butyrate-producing bacteria reduction. Our previous study suggested administration of Clostridium butyricum (C. butyricum), a bacterium, exerts crucial effect against CRC. Methyltransferase-like 3 (METTL3) contributes tumorigenic epigenetic regulation. We aimed investigate the effects C. on METTL3 in prevention Methods TCGA and CancerSEA databases were used co-expressed genes, GO KEGG enrichment analysis was conducted METTL3. Tissue specimens human normal colonic tissue, adenoma carcinoma obtained examined expression METTL3, EMT-associated markers, VM formation. overexpressed CRC cells assess cell migration tube Meanwhile, epithelial-mesenchymal transition (EMT), vasculogenic mimicry (VM) formation evaluated. Furthermore, murine xenograft model established provide further evidence inhibition tumor growth. Results Database that showed positive correlation with proliferation, EMT, DNA repair, metastasis, invasion. The gradually increased from colon carcinoma, positively correlated EMT. overexpression promoted migration, invasion induced could downregulate decrease vimentin vascular endothelial growth factor receptor 2 reduce EMT Moreover, alleviated pro-oncogenic overexpressing plasmid cells. Accordingly, downregulated tumors prevented nude mice. Conclusion inhibit intestinal carcinogenesis through downregulating These findings broaden our understanding probiotics supplement treatment

Casticin inhibits epithelial-mesenchymal transition of liver cancer stem cells of the SMMC-7721 cell line through downregulating Twist

The existence of cancer stem cells (CSCs) is central to the pathogenesis and therapeutic target of human hepatocellular carcinoma. The aim of this study was to investigate the effects of casticin on epithelial-mesenchymal transition (EMT) of liver cancer stem cells (LCSCs) derived from the SMMC-7721 cell line. Our results demonstrated that CD133+ sphere-forming cells (SFCs) sorted from the SMMC...

Thymoquinone inhibits cancer metastasis by downregulating TWIST1 expression to reduce epithelial to mesenchymal transition

Proteins that promote epithelial to mesenchymal transition (EMT) are associated with cancer metastasis. Inhibition of EMT regulators may be a promising approach in cancer therapy. In this study, Thymoquinone (TQ) was used to treat cancer cell lines to investigate its effects on EMT-regulatory proteins and cancer metastasis. We show that TQ inhibited cancer cell growth, migration and invasion in...

miR-671-5p inhibits epithelial-to-mesenchymal transition by downregulating FOXM1 expression in breast cancer.

MicroRNA (miRNA) dysfunction is associated with a variety of human diseases, including cancer. Our previous study showed that miR-671-5p was deregulated throughout breast cancer progression. Here, we report for the first time that miR-671-5p is a tumor-suppressor miRNA in breast tumorigenesis. We found that expression of miR-671-5p was decreased significantly in invasive ductal carcinoma (IDC) ...

N-acetylcysteine inhibits alveolar epithelial-mesenchymal transition.

The ability of transforming growth factor-beta1 (TGF-beta1) to induce epithelial-mesenchymal transition (EMT) in alveolar epithelial cells (AEC) in vitro and in vivo, together with the demonstration of EMT in biopsies of idiopathic pulmonary fibrosis (IPF) patients, suggests a role for TGF-beta1-induced EMT in disease pathogenesis. We investigated the effects of N-acetylcysteine (NAC) on TGF-be...

53BP1 suppresses epithelial–mesenchymal transition by downregulating ZEB1 through microRNA-200b/429 in breast cancer

Epithelial-mesenchymal transition (EMT) is an important mechanism of cancer invasion and metastasis. Although p53 binding protein 1 (53BP1) has been implicated in several biological processes, its function in EMT of human cancers has not yet been reported. Here, we show that 53BP1 negatively regulated EMT by modulating ZEB1 through targeting microRNA (miR)-200b and miR-429. Furthermore, 53BP1 p...
